Abstract

La recerca recollida en aquest manuscrit es centra en els metal·lacarborans cobalta i ferrabisdicarballur ([3,3’-Co(1,2-C2B9H11)2]-, [1]-; i [3,3’-Fe(1,2-C2B9H11)2]-, [2]-, respectivament), compostos aniònics i redox reversible, emprats com a punt de partida i objectiu final. El primer capítol de la Tesi es basa en la cerca d’una estratègia per funcionalitzar regioselectivament una plataforma simple amb diferents tipus de metal·lacarborans ([1]- i [2]-), amb bons rendiments i de fàcil purificació. Per assolir aquest objectiu, s’ha optat per una ruta sintètica neta, amb bons rendiments i eficient: la síntesi malònica i l’acetoacètica. Seguint aquest procediment, s’ha obtingut l’homo i heterosubstitució, obtenint per primera vegada una plataforma heterofuncionalitzada amb [1]- and [2]-, amb alts rendiments. El segon apartat de la Tesi es refereix a les propietats electroquímiques dels metal·lacarborans. La determinació i modulació del potencial redox de metal·lacarborans funcionalitzats, mitjançant la voltamperometria cíclica, ha estat abastament estudiada aquí. El E1/2 de la plataforma de metal·lacarborà ha estat eixamplat fins a 2 V per primera vegada, mitjançant la iodació seqüencial dels seus vèrtex B-H. A més a més, s’han fet experiments d’electròlisi per intentar entendre i demostrar la reversibilitat i estabilitat dels productes oxidats i reduïts generats, respectivament. Addicionalment, s’ha vist que els metal·lacarborans enllaçats iònicament amb el metil viologen són bons materials polielectrocròmics. Finalment, un últim estudi d’aquest mateix capítol es centra en la capacitat d’inhibició de fluorescència de fluoròfors per part dels metal·lacarborans, així com la influència de la càrrega, E1/2 i configuració electrònica d’aquests en l’eficiència d’inhibició final. La tercera part tracta les interaccions i afinitats iòniques dels metal·lacarborans en solució aquosa. Es centra en molècules amb dues unitats de metal·lacarborà connectades per diferents cadenes polietoxilades (tipus pesa de gimnàs), i la seva habilitat “d’abraçar” diferents cations. En aquest estudi s’han estudiat cations alcalins, essent els de major radi (K+ i Cs+) els més afavorits i els que millor encaixen en els requeriments electrònics i estèrics dels diferents compostos estudiats; mentre que els de menor radi (Li+ i Na+) es veuen clarament desafavorits. Per últim, el quart capítol tracta les interaccions de [1]- i els seu derivat diiodat, [32]-, amb cèl·lules vives. Tenint present la solubilitat en medi aquós i les interaccions d’aquests tipus de compostos, s’han fet estudis de biologia molecular amb cèl·lules de mamífers, amebes i bacteris. Els primers resultats confirmen la penetració dels metal·lacarborans a l’interior de les cèl·lules, produint un arrodoniment instantani de la forma cel·lular. La distribució d’aquest compost a l’interior de les cèl·lules vives és heterogènia, suggerint algun tipus d’interacció amb part específiques de l’interior. Sorprenentment, les cèl·lules en contacte amb els compostos de bor aturen la seva activitat de divisió cel·lular, recuperant-la un cop s’extreu el compost del medi. A més a més, altes concentracions dels metal·lacarborans no maten les cèl·lules, confirmant que aquests no són directament citotòxics. També s’ha demostrat que l’efecte citostàtic és més o menys efectiu en funció del tipus cel·lular estudiat, essent [32]- més potent/eficient que [1]-, en tots els casos. Finalment, no s’han observat modificacions o degradacions químiques dels metal·lacarborans emprats, podent recuperar els compostos de bor al final dels experiments, habilitant-los per a posteriors usos.


The work presented in this manuscript deals with cobalt and ferrabisdicarbollide metallacarboranes ([3,3’-Co(1,2-C2B9H11)2]-, [1]-; and [3,3’-Fe(1,2-C2B9H11)2]-, [2]-, respectively), anionic and redox reversible compounds, used as starting point and final target. The first section of the Thesis is focused on the search for an efficient strategy to regioselectively functionalize a simple platform with different kind of metallacarboranes ([1]- and [2]-), with good yields and easy purification processes. A very clean, high-yield and efficient synthetic pathway has been found: the malonic and acetoacetic ester synthesis. Following this synthetic route, the homo and heterosubstitution has been obtained, achieving for the first time the heterofunctionalization of a single platform with [1]- and [2]-, with high yields. The second chapter of the Thesis is referred to the electrochemical properties of metallacarboranes. The redox potential determination and modulation of the differently substituted metallacarboranes, by means of cyclic voltammetry, has been widely studied here. The E1/2 of the metallacarborane platform has been broadened up to 2 V for the first time, by sequential iodinations of its B-H vertexes. In addition, electrolysis experiments have been carried out, trying to understand and demonstrate the reversibility and stability of the generated reduced and oxidized products, respectively. Moreover, they turned to be good polyelectrochromic materials when coupled with methyl viologen. Furthermore, a last study in this same chapter demonstrates the fluorescence quenching ability of metallacarboranes, and the influence of the charge, E1/2 and electronic configuration of these boron cluster compounds in terms of quenching efficiency. The third section is related to interactions and ionic affinities of metallacarboranes in aqueous solution. This chapter is focused on double metallacarborane molecules connected by different polyethoxylated chains (dumbbell-like), and their ability to “embrace” different cations. In this study, alkaline cations have been tested, being the larger ones (K+ and Cs+) the preferred and the ones that better fit the electronic and steric requirements of the different metallacarborane-dumbbells; while the smaller ones (Li+ and Na+) are clearly less favored. At last, but certainly not least, the fourth chapter deals with the interactions of [1]- and its diiodinated derivative, [32]-, with living cells. Bearing in mind the water solubility and intermolecular interactions of such compounds, molecular biology studies with mammalian cells, amoebas and bacteria have been carried out. The first results confirmed the penetration of metallacarboranes inside living cells, producing an immediate rounding of the cell shape. The distribution of the boron-based compounds inside the cells turned to be heterogenic, suggesting some kind of interaction with specific internal parts of the cells. Strikingly, the cells in touch with the boron compounds stopped their cell division activity, and recovered it once the compound is removed from the medium. Additionally, high loads of the metallacarboranes do not kill cells, confirming that the clusters are not directly toxic. It has also been demonstrated that the citostatic effect is more or less effective depending on the studied cell, but in all cases [32]- turned to be more potent/efficient than [1]-. Besides, no chemical modifications or degradations have been observed on the used metallacarboranes, recovering the original compounds at the end of the experiments, enabling them for further uses.
