The source of interleukin-6 in the central nervous system determines its effects on body weight and behavior

Abstract

La interleucina-6 (IL-6) es una citocina pleiotrópica con funciones tanto inflamatorias como no inflamatorias. Participa en la diferenciación de células B a células plasmáticas, la inducción de proteínas de fase aguda y la regulación de la transición entre la inmunidad innata y adquirida. En cuanto a sus funciones no relacionadas con la inflamación, participa en la regulación del peso corporal y el comportamiento, entre otras. Dado que es producida por múltiples tipos celulares, tanto en la periferia como en el sistema nervioso central (SNC), es clave determinar el papel de cada una de las fuentes y las células en las que actúa. Con este fin, hemos generado ratones knock-out condicionales para la IL-6 en astrocitos o neuronas, así como ratones knock-out condicionales para el receptor de la IL-6 (IL-6R) en neuronas, para estudiar la influencia de esta citocina en la regulación del peso corporal en situación basal y tras diferentes desafíos calóricos, como una dieta rica en grasas o el ayuno. También hemos empleado ratones con expresión crónica de IL-6 en astrocitos, ratones GFAP-IL6, con el fin de ampliar nuestro estudio del efecto central de la IL-6. Cabe mencionar, que nos encontramos una recombinación del gen de interés fuera de las células diana debido a actividad Cre recombinasa no específica, que fue más prominente con el promotor neuronal (Syn1) que con el astrocitario (GFAP). Este punto es muy relevante y debe ser tenido en cuenta al trabajar con líneas de ratones que expresen Cre. Nuestros resultados muestran, por una parte, que tanto la IL-6 astrocitaria como la neuronal fomentan la supervivencia in utero, restringen la exploración y facilitan el comportamiento de tipo ansioso. Por otra parte, cada IL-6 tiene efectos opuestos en la ganancia de peso corporal con una dieta rica en grasas, inhibiéndola la IL-6 astrocitaria y favoreciéndola la neuronal; en el crecimiento longitudinal, en el que la IL-6 astrocitaria lo inhibe mientras que la neuronal lo promueve; y en la locomoción general, que está facilitada por la IL-6 astrocitaria y lo contrario por la neuronal. Además, nuestros resultados con los ratones knock-out condicionales para el IL-6R neuronal indican que los efectos de la IL-6 neuronal en el crecimiento longitudinal y la ingesta compensatoria tras un ayuno son autocrinos, ya que ambos modelos presentan fenotipos similares. Por el contrario, el receptor neuronal no parece mediar los efectos en el comportamiento. En resumen, la célula productora de IL-6 en el SNC es crítica para determinar sus efectos sobre diversas variables relacionadas con el peso corporal y el comportamiento en el ratón.


Interleukin-6 (IL-6) is a pleiotropic cytokine with inflammatory and non-inflammatory effects. It participates in the differentiation of B-cells into high-rate Ig-secreting cells, the induction of acute phase response proteins and the orchestration of the transition from innate to acquired immunity. Regarding its non-inflammation-related roles, it is involved in the regulation of body weight and behavior, among others. Since it can be produced by numerous cell types, both in the periphery and the central nervous system (CNS), it is of paramount interest to ascertain the role of each source and the cells on which it acts. To this end, we have generated conditional IL-6 knock-out mice in neurons and astrocytes, as well as conditional IL-6 receptor (IL-6R) knock-out mice in neurons to study the influence of this cytokine in the regulation of body weight in basal conditions and after different caloric challenges, such as a high-fat diet (HFD) and fasting. We have also used mice with chronic expression of IL-6 in astrocytes, GFAP-IL-6 mice, to gain more insight into the central effect of IL-6. Of note, we have encountered extensive off-target recombination of our genes of interest due to leaked Cre recombinase activity, more prominent with the neuronal promoter, Syn1, than with the astrocytic, GFAP. This is a very relevant point to take into account when working with Cre driver lines. Our results show that, on the one hand, both astrocytic and neuronal IL-6 have a pro-survival effect in utero, similarly restrict exploration and facilitate anxiety-like behavior. And, on the other hand, they have opposite effects on body weight gain after a HFD, with astrocytic IL-6 preventing obesity and neuronal IL-6 promoting it; on longitudinal growth, where astrocytic IL-6 inhibits it and neuronal IL-6 promotes it; and on general locomotion, which is facilitated by astrocytic IL-6 and inhibited by neuronal IL-6. Also, our results with conditional IL-6R knock-out mice indicate that the effects of neuronal IL-6 on longitudinal growth and compensatory food intake after a fast are autocrine, since both models display similar phenotypes. Conversely, the neuronal receptor does not seem to mediate the effects on behavior. In summary, the CNS cell producing IL-6 is critical in determining the effect on several variables related to body weight and behavior in the mouse.
