Abstract

La Tesi tracta sobre la preparació i l’ús en catàlisi homogènia asimètrica de lligands fosforats quirals amb àtoms de fòsfor estereogènics. En primer lloc es descriu la preparació i caracterització de lligands monofosforats P-estereogènics diarílics amb un substituent policíclic o heterocíclic, de fórmula general PArPhR’, on Ar = 2-p-terfenilil, 1-pirenil, 4-dibenzofuril, 4-dibenzotiofenil o 1-tiantrenil i R = OMe, Me o i-Pr, t-Bu o Fc. El mètode sintètic és el de Jugé-Stephan, en el qual s’empra l’aminoalcohol efedrina com a auxiliar quiral. Els lligands (P*) s’han coordinat a fragments organometàl·lics de Pd i Ru per conduir als complexos de tipus [Pd(al·lil)Cl(P*)], [Pd(al·lil)(k2P,S-P*)]PF6 [Pd(al·lil)(P*)2]PF6, [Ru(arè)Cl2(P*)] i [Ru(arè)Cl(k2P,S-P*)]PF6. Els complexos s’han caracteritzat en estat sòlid i en solució i s’han utilitzat com a precursors catalítics en la hidrovinilació asimètrica de l’estirè catalitzada per Pd, on s’han obtingut altes activitats (amb TOFs de fins 1839 h–1) i altes selectivitats (> 90% cap a 3-fenil-1-butè) en alguns casos. L’alquilació al·lílica asimètrica de l’acetat d’1,3-difenilal·lil catalitzada per Pd ha conduït a conversions completes a 24 hores i fins un 80% d’excés enantiomèric. En la hidrogenació asimètrica de l’acetofenona per transferència d’hidrogen catalitzada per Ru s’ha arribat a conversió completa a 3 hores en alguns casos, i a valors d’excés enantiomèric de fins un 70%. A la segona part es descriu la preparació de lligands difosforats amb simetria C1 que contenen un fragment fosfina, en la majoria de casos P-estereogènica, i un altre fragment de tipus fosfinit aquiral, fosfit quiral o diamidofosfit quiral. Els lligands s’han obtingut per condensació entre un fosfinoalcohol i un precursor clorofosforat en presència de base. Els lligands (P’-P*; P’ = fosfinit, fosfit o diamidofosfit; P* = fosfina) s’han coordinat de forma bidentada a Pd i Ru i s’han obtingut compostos del tipus [Pd(al·lil)(P’- P*)]PF6 i [Ru(arè)Cl(P’-P*)]PF6. Després de caracteritzar-los, els complexos s’han emprat en la substitució al·lílica de diversos substrats i s’han aconseguit excel·lents enantioselectivitats, de fins el 96% d’ee. La configuració absoluta del producte majoritari s’ha racionalitzat mitjançant càlculs teòrics. Els complexos de Ru també s’han emprat en la reducció de l’acetofenona i s’han assolit conversions completes a 1 h i un 30% d’ee.


The present Thesis focusses on the preparation of chiral phosphorus ligands containing stereogenic phosphorous atoms and their use in asymmetric homogeneous catalysis. In the first part, the synthesis and characterisation of new P-stereogenic diarylic monophosphorate ligands containing polycyclic or heterocyclic substituents is described. The general formula of these kind of compounds is (S)-PPhRR’, where Ar = 2-p-terphenylyl, 1-pirenyl, 4-dibenzofuranyl, 4- dibenzotiophenyl or 1-thiantrenyl and R = OMe, Me or i-Pr, t-Bu or Fc. The synthetic procedure is similar to Jugé-Stephan method, based on the use of ephedrine as chiral auxiliary. These ligands (P*) have been coordinated to Pd and Ru organometallic moieties in order to prepare complexes with the general formula [Pd(allyl)Cl(P*)], [Pd(allyl)(k2P,S-P*)], PF6 [Pd(allyl)(P*)2]PF6, [Ru(arene)Cl2(P*)] and [Ru(arene)Cl(k2P,S-P*)]PF6. The corresponding complexes have been characterized both in solution and in the solid state. In addition, Pd complexes have been tested as catalytic precursors in the Pd catalyses asymmetric hydrovinylation of styrene, obtaining good activities (TOFs up to 1839 h–1) and selectivities (> 90% to 3-phenyl-1-butene) as well. In the Pd catalyses asymmetric allylic alkylation reaction of 1,3-diphenylallyl acetate full conversion is achieved at 24 hours with high enantioselectivity, at least 80% ee. Furthermore, we have observed, in all cases, full conversion at 3 hours and moderate enantioselectivity (70% ee) in the Ru catalyses asymmetric hydrogen transfer reaction using acetophenone as substrate. The second part is dealt on the preparation and characterisation of diphosphorate ligands with C1 symmetry, containing a chiral phosphine moiety, in most cases P-stereogenic, while the other fragment is an aquiral phosphinite or a quiral phosphite or diamidophosphite respectively. All these ligands have been prepared by condensation of a phosphinoalcohol with a clorophosphorate reagent in presence of a base. The bidentate ligands (P’-P*; P’ = phosphinite, phosphite o diamidophosphite; P* = phosphine) have also been coordinated to both, Pd and Ru metallic moieties, in order to obtain compounds with the formula [Pd(allyl)(P’-P*)]PF6 and [Ru(arene)Cl(P’-P*)]PF6. After their characterization, these complexes have been used in the asymmetric allylic substitution reaction in presence of different substrates achieving excellent enantioselectivities (at least 96% ee). The absolute configuration of the major enantiomer have been rationalized by theoretical calculations. The Ru complexes have been used in the asymmetric hydrogen transfer of acetophenone showing full conversions to 1 h with 30% of ee.
