The aim of the thesis "Evaluation of the neurobehavioral effects of manganese exposure on rodents. Influence of stress", is to study and deepen about the manganese (Mn) potential prenatal, postnatal and adult toxic effects on the Central Nervous System (CNS), and about the possible impact of the stress situation on these toxic effects.<br/>Mn, as other trace elements, must be between certain limits of concentration in human and animal bodies, to work on well. An over exposition to Mn is associated to changes on CNS, with psychologic and neurologic symptoms, at the beginning the features are psychiatric alterations ("manganese madness"), that are very similar to the esquizofrenic ones. While at the first stages it can revert, at he last stages it persists, being irreversible.<br/>Development period is considered especially susceptible to a wide range of toxic expositions. Young animals not only absorve and retention Mn dramatically, but also they had showed a higher relative accumulation of Mn in CNS.<br/>Research in neurotoxicology on human beings give us advantages and disadvantages. In epidemiologic studies cause-effect or dose-effect relations can not be detected. Experimental control over the exposition is required to identify the relation between dose and Mn derived effects, in order to avoid this we chose research animals, rodents, to work on them. <br/>Mn neurotoxicity has been demonstrated when it is inhaled or injected, while possible toxic effects from Mn oral ingestion has been poorly studied, and data reported are not conclusive. There is also a lack of attention on Mn effects on development.<br/>After Mn intoxication symptoms observed in human beings are resembling to Parkinson symptoms, and this coincidence force the research to dopaminergic system, which has been extensively studied in relation to Mn effects. However, nowadays, the study of possible mechanisms of Mn neurotoxic action not only involves dopaminergic system, but also other transmitter systems. On the other hand, physic and psychologic stressors, provoke changes at CNS and more specifically, at dopaminergic system level.<br/>Given that, Mn and stressful situations affect, directly or indirectly, dopaminergic system function, Mn exposition at the same time that stress exposition could result as an interaction, increasing or modifying the effects of Mn on the CNS. The aim of this thesis is to assess the neurobehavioral effects of Mn prenatal, postnatal and in adults exposure on rodents and interactions of Mn and stress.<br/>Four studies has being done to reach the general goal of this thesis; and in with each one we could assess:<br/> Materno-toxic and feto-toxic effects of hydrocortisone (HC) and Mn prenatal exposure.<br/> Effects of Mn and stress prenatally exposed on postnatal development.<br/> Effects of chronic Mn oral exposure and stress exposure in adults.<br/> Neurobehavioral effects of two kinds of stress: restraint and noise.<br/>All these assessments gave us data about Mn toxicity: maternal, fetal, developmental and in adult animals. We also evaluated two types of stress models because of the rising interest in the assessment of stressful situations and more specifically in order to know the most suitable model of stress to use, in each case, in the future.
La tesi doctoral que porta per títol "Avaluació dels efectes neuroconductuals de l'exposició a manganès en animals d'experimentació. Influència de l'estrès", vol estudiar i aprofundir en els potencials efectes tòxics del manganès (Mn) sobre el sistema nerviós central (SNC), i la possible influència que les situacions d'estrès exercirien sobre aquesta toxicitat, a nivell prenatal, postnatal i en els éssers ja adults.<br/>El Mn, com altres elements essencials, ha de mantenir-se entre uns certs límits de concentració per un funcionament òptim de l'organisme. L'exposició excessiva a Mn està associada amb canvis al SNC, amb símptomes tant psicològics com neurològics, caracteritzats en el seu inici per una alteració psiquiàtrica ("bogeria mangànica") que recordaria força a l'esquizofrènia. Mentre que en fases inicials pot ser reversible, el manganisme més avançat persistiria, essent irreversible.<br/>El període del desenvolupament es considera especialment susceptible a un ampli rang d'exposicions tòxiques. Els animals molt joves no només absorbeixen i retenen Mn de manera excepcional sinó que també han mostrat una acumulació relativa més gran del Mn al SNC.<br/>Els estudis de neurotoxicitat del Mn conduïts en poblacions humanes comporten avantatges i desavantatges. En els estudis epidemiològics, les relacions causa-efecte o dosi-efecte no poden ser identificades. Es requereix el control experimental sobre l'exposició per poder identificar clarament relacions entre la dosi i l'efecte del Mn, d'aquí l'elecció de treballar amb animals d'experimentació, rosegadors.<br/>La neurotoxicitat del Mn ha estat demostrada quan aquest és inhalat o bé administrat per via parenteral. En canvi, els possibles efectes derivats del consum elevat de Mn per via oral han estat poc estudiats i no existeixen dades concloents al respecte. Així mateix, els efectes del Mn sobre el desenvolupament han rebut poca atenció.<br/>La simptomatologia observada en humans després de la intoxicació per Mn és semblant a la observada en la malaltia de Parkinson, la qual cosa ha fet que el sistema dopaminèrgic hagi estat àmpliament estudiat en relació als efectes del Mn. Tot i que en l'actualitat els possibles mecanismes de l'acció neurotòxica del Mn no es centren només en aquest sistema. Per altra banda, els estressors tant físics com psicològics, provoquen canvis a nivell de SNC, i concretament a nivell del sistema dopaminèrgic.<br/>Donat que el Mn i l'estrès afecten de forma directa o indirecta el funcionament del sistema dopaminèrgic, l'exposició concurrent a Mn i estrès es pot manifestar com una interacció, augmentant o modificant els seus efectes sobre el SNC. L'objectiu general d'aquest treball ha estat dirigit a avaluar els efectes neuroconductuals de l'exposició a Mn en rosegadors sobre la toxicitat materna, fetal, postnatal i en el subjecte adult, així com la influència que l'estrès pot exercir en aquesta exposició. <br/>En concret s'han fet 4 estudis per aconseguir l'objectiu general d'aquesta tesi; i en cadascun d'ells s'han estudiat:<br/>Els efectes materno i feto-tòxics de l'exposició prenatal a hidrocortisona (HC) i manganès (Mn), administrats s.c. (subcutani)<br/> Els efectes de l'exposició prenatal a Mn (administrat s.c.) i estrès en el desenvolupament postnatal.<br/> Els efectes de l'exposició crònica en adults a Mn i estrès, administrat via oral.<br/> Els efectes neuroconductuals de dos tipus d'estrès: estrès per immobilització i estrès per soroll.<br/>Mitjançant aquests estudis s'han obtinguts resultats que ens donen informació sobre la toxicitat: materna, fetal, durant el desenvolupament i en els animals adults. També hem avaluat diferents tipus o models d'estrès degut al gran interès que té l'avaluació de l'estrès i, en concret, la idoneïtat del model d'estrès a utilitzar en cada cas i en el futur.
