Los factores guía son proteínas esenciales en el proceso de guía de neuronas y axones que ocurre durante el desarrollo del sistema nervioso. Netrinas y Semaforinas componen dos de las cinco familias de factores guía existentes. En este trabajo, nosotros hemos estudiado el efecto quimiotáctico de Netrina1 y Semaforinas secretables Sema3A y Sema3F en la guía de interneuronas y sus axones durante el desarrollo del cerebelo y el hipocampo, así como la participación de Sema3A en el proceso de regeneración de axones axotomizados en un modelo in vitro. <br/><br/>El primer objetivo de nuestro trabajo fue el estudio de la función de Netrina1 en el desarrollo postnatal de interneuronas de la corteza cerebelar, y pudimos constatar que: <br/><br/>a) Netrina1 es un factor quimiorepulsivo in vitro para interneuronas de la corteza cerebelar postnatal; <br/><br/>b) Netrina1 y sus receptores Dcc, Unc5h2 y Unc5h3 se expresan en la corteza cerebelar durante el desarrollo postnatal; <br/><br/>c) las interneuronas de la corteza postnatal cerebelar expresan Netrina1 y sus receptores Dcc, Unc5h2 y Unc5h3; <br/><br/>d) Dcc no parece participar en la señal quimiorepulsiva de Netrina1 ante interneuronas de la corteza cerebelar; <br/><br/>e) las interneuronas de la corteza cerebelar migran tanto in vitro como in vivo asociadas a fibras gliales. <br/><br/>Nuestro segundo objetivo fue estudiar el efecto de Sema3A y Sema3F sobre axones GABAérgicos de la formación hipocampal en desarrollo. Hemos observado que: <br/><br/>a) Sema3A y Sema3F son agentes quimiorepulsivos in vitro sobre axones hipocampales embrionarios y postnatales, mientras que solamente Sema3F repele axones entorrinales embrionarios; <br/><br/>b) Sema3A, Sema3F y sus respectivos receptores Neuropilina1 y Neuropilina2 se expresan en el hipocampo a edades embrionarias y postnatales; <br/><br/>c) interneuronas GABAérgicas de la capa piramidal y el estrato oriens expresan Neuropilina1 y 2, mientras que aquellas situadas en los estratos radiado y lacunosum-moleculare no expresan dichos receptores. <br/><br/>Nuestro tercer y último objetivo fue estudiar el posible efecto inhibitorio de Sema3A sobre axones adultos axotomizados, utilizando el cultivo organotípico entorrino-hipocampal como modelo de lesión in vitro. Nosotros demostramos que: <br/><br/>a) tras axotomía a 15 días in vitro (DIV), cuando los axones entorrinales no regeneran, aumenta la expresión de Sema3A tanto en hipocampo como en corteza entorrinal, y también aumenta la expresión de su receptor Neuropilina1 en corteza entorrinal; <br/><br/>b) en cultivos organotípicos entorrino-hipocampales obtenidos de ratones homocigotos deficientes en Sema3A, se produce la regeneración parcial de axones entorrinales axotomizados a 15 DIV.
<I>Guidance cues are essential during the development of the central nervous system. In the present Thesis, we studied the migration of postnatal interneurons of the cerebellar cortex, and the role of Netrin1 in this process; furthermore, we studied the effect of Sema3A and Sema3F in the extension of GABAergic axons projecting locally in the development of hippocampal formation, and the role of Sema3A in the regeneration process of adult injured axons using the in vitro model of the entorhino-hippocampal organotypic culture. We obtained the following results:<br/><br/>- Interneurons of the postnatal cerebellar cortex migrate closely associated to glial fibres, both in-vivo and in-vitro;<br/><br/>- Netrin1 is a chemorepulsive cue for interneurons of the postnatal cerebellar cortex in vitro, and the expression pattern of Netrin1 and its receptors Dcc, Unc5h2 and Unc5h3 as well as the fact that interneurons express these receptors in vivo, indicate that Netrin1 may influence the migration of interneurons in vivo.<br/><br/>- Dcc does not participate in the repulsive signal that Netrin1 exerts on interneurons of the cerebellar cortex in vitro.<br/><br/>- Sema3A and Sema3F have a chemorepulsive effect on GABAergic axons from the embryonic and postnatal hippocampus, but only Sema3F is a chemorepulsive cue for GABAergic axons from the embryonic entorhinal cortex.<br/><br/>- The expression pattern of Sema3A, Sema3F and their respective receptors Neuropilin1 and 2, and the fact that some of the GABAergic interneurons in the embryonic and postnatal hippocampus express those receptors, suggest a possible role of Sema3A and Sema3F in the axonal development of hippocampal GABAergic interneurons.<br/><br/>- The axotomy of the entorhino-hippocampal pathway increases the expression of Sema3A and its receptor Neuropilin1 in entorhino-hippocampal organotypic cultures.<br/><br/>- The lack of Sema3A in entorhino-hippocampal organotypic cultures yields a partial regeneration of injured entorhinal axons. </i>
