La leucemia aguda es una alterción clonal de los progenitores hematopoyéticos, con una gran heterogeneidad tando desde el punto de vista clínico como biológico. Actualmente el tratamiento de los pacientes con leucemia aguda se estratifica según factores pronósticos. Disponemos de diversas técnicas útiles en el diagnóstico, clasificación y seguimiento de los enfermos con leucemia aguda y es necesario integrar todas ellas para conseguir una correcta caracterización de la leucemia aguda. De esta manera se definen, dentro de la heterogeneidad de la leucemia aguda, entidades clínico-biológicas con determinado valor pronóstico y tributarias a un tratamiento más individualizado. En este sentido, el análisis inmunofenotípico en la leucemia aguda es imprescindible para la correcta clasificación, aporta información pronóstica y más recientemente se utiliza en el estudio de la enfermedad residual mínima en la remisión.<br/>El objetivo de esta tesis es analizar el valor en la caracterización de la leucemia aguda de nuevos marcadores como CD66 y CD123 y estabecer correlaciones entre el patrón inmunofenotípico y la presencia de lesiones moleculares con valor pronóstico como los reordenamientos: bcr/abl en la LLA y aml-1/eto, cbfb/myh-11, mll y duplicación de flt3 en la LMA.Acute lekemias are considered to be clonal disorders involving early hematopoietic progenitor cells, with a wide clinical and biological heterogeneity. Clinical outcome and treatment depends on prognostic factors. Actually, correlation of several techniques is mandatory for the correct knowledge of this disease. In this way, phenotype analysis by flow cytometry is usful for acute leukemia classification, and more recently, for the study of minimal residual disease. <br/>The aim of this thesis is 1) To analyze the value new markers such us: CD66 and CD123 in order to distinguish better the phenotypic properties of normal and tumoral cells and their utility in the study of minimal residual disease and 2) To analyze the utility of immunophenotype for the screening of specific genetic lesions common in acute leukemia such us: BCR/ABLl in ALL and AML-1/ETO, CBFB/MYH-11, MLL rearrangements and FLT3 mutations in AML.
