Este trabajo de tesis presenta la caracterización del efecto anticanceroso y la identificación de dianas moleculares de principios activos procedentes de la bacteria "Serratia marcescens". En particular, se ha evaluado la capacidad anticancerosa "in vitro" y "ex vivo" del ciclodepsipéptido serratamolide (AT514) en varias células cancerosas y no cancerosas de diverso origen tisular, obteniendo una mayor sensibilidad al tratamiento por parte de las células cancerosas. También hemos realizado estudios para evaluar la toxicidad "in vivo" de AT514. Respecto al tripirrol prodigiosina, hemos ampliado los conocimientos acerca de su propiedad anticancerosa evaluando células de cáncer de mama, pulmón y neuroblastoma. En todos los casos se ha obtenido un marcado descenso de la viabilidad celular, incluso en células con fenotipo de resistencia a múltiples fármacos, demostrando que prodigiosina no es sustrato de algunas de las proteínas ABC responsables de dicho fenómeno. Hemos determinado que ambas sustancias son capaces de ejercer un efecto citostático o citotóxico según el tipo celular, dosis y tiempo de exposición al tratamiento. <br/>También se ha analizado el mecanismo de acción por el cual ejercen su actividad anticancerosa. AT514 interacciona con la vía de proliferación PI3K/AKT/NF-kB, provocando desfosforilación de la quinasa AKT e inhibiendo la actividad transcripcional de NF-kB. Mediante el análisis de las variaciones en la expresión génica y en el perfil proteómico tras el tratamiento con prodigiosina, hemos identificado una serie de genes diana. Entre las proteínas que más modificaron su expresión encontramos algunas relacionadas con detoxificación, reparación de ADN y proteínas estructurales. En los experimentos de macroarrays identificamos 37 genes que modulaban su expresión de forma significativa, de los cuales el gen antitumorigénico NAG-1 fue el más modulado. Tras el tratamiento con prodigiosina, dicho gen es inducido por la quinasa GSK-3beta, la cual demostramos que tiene un papel crucial en los efectos producidos por la droga, ya que induce la expresión de los receptores de muerte DR-4 y -5, así como la del inhibidor del ciclo celular p21. También hemos observado que la activación de la vía de la citoquina TGF-beta es necesaria para la inducción de p21. Asimismo, el efecto anticanceroso de ambas sustancias es independiente del estado funcional del gen supresor de tumores p53, propiedad beneficiosa ya que éste se encuentra mutado en la mayoría de tumores humanos. <br/>Los resultados obtenidos muestran detalladamente las rutas moleculares que activan dos nuevas sustancias anticancerosas presentándolas como potenciales agentes quimioterapéuticos.
This works shows the characterization of the anticancerous effect and the identification of the molecular targets of bioactive molecules from the bacteria Serratia marcescens. In particular, we have studied the anticancerous ability in vitro and ex vivo of the cyclodepsipeptide serratamolide (AT514) in several cancerous and non-cancerous cells from different origin. We have also evaluated the in vivo toxicity of AT514. Regarding the tripirrol prodigiosin, we have extended our knowledge about its anticancerous ability evaluating breast, lung and neuroblastoma cancer cells, including some multidrug resistant cancer cell lines. Both molecules induce citostatic or citotoxic effects depending on the cellular type, dose and drug exposure time, with more potent effects in cancerous cells. <br/>We have also studied the mechanism of action that they activate in order to trigger their anticancerous effect. AT514 interacts with the prosurvival pathway PI3K/AKT/NF-¦ÊB, inducing dephosphorilation of the kinase AKT and inhibiting the transcriptional activity of NF-¦ÊB. Through the analysis of variations in gene and protein expression after treatment with prodigiosin, we have identified several target genes. Among the proteins that significantly modified their expression, we found some related to cellular detoxification, DNA repair and structural proteins. In the macroarrays experiments, we identified 37 genes that significantly modulated their expression, being the antitumorigenic gene NAG-1 the most modulated one. After prodigiosin treatment, GSK-3¦Â induced the expression of this gene. This kinase has a key role in the effects provoked by this drug as it induced also death receptor 4 and 5 (DR-4, -5) and cell cycle inhibitor p21 gene expression. We have also observed that activation of the citokine TGF-¦Â pathway is necessary for p21 induction. Moreover, the anticancerous effect of both substances is independent of the tumor supressor gene p53 functional status, advantageous trait as it is mutated in the vast majority of human tumours.<br/>The results obtained show in detail the molecular pathways that are activated by two novel anticancerous agents, pointing them as potential chemotherapeutic agents.
