The aim of the thesis "Evaluation of the neurobehavioral effects of manganese exposure on rodents. Influence of stress", is to study and deepen about the manganese (Mn) potential prenatal, postnatal and adult toxic effects on the Central Nervous System (CNS), and about the possible impact of the stress situation on these toxic effects.<br/>Mn, as other trace elements, must be between certain limits of concentration in human and animal bodies, to work on well. An over exposition to Mn is associated to changes on CNS, with psychologic and neurologic symptoms, at the beginning the features are psychiatric alterations ("manganese madness"), that are very similar to the esquizofrenic ones. While at the first stages it can revert, at he last stages it persists, being irreversible.<br/>Development period is considered especially susceptible to a wide range of toxic expositions. Young animals not only absorve and retention Mn dramatically, but also they had showed a higher relative accumulation of Mn in CNS.<br/>Research in neurotoxicology on human beings give us advantages and disadvantages. In epidemiologic studies cause-effect or dose-effect relations can not be detected. Experimental control over the exposition is required to identify the relation between dose and Mn derived effects, in order to avoid this we chose research animals, rodents, to work on them. <br/>Mn neurotoxicity has been demonstrated when it is inhaled or injected, while possible toxic effects from Mn oral ingestion has been poorly studied, and data reported are not conclusive. There is also a lack of attention on Mn effects on development.<br/>After Mn intoxication symptoms observed in human beings are resembling to Parkinson symptoms, and this coincidence force the research to dopaminergic system, which has been extensively studied in relation to Mn effects. However, nowadays, the study of possible mechanisms of Mn neurotoxic action not only involves dopaminergic system, but also other transmitter systems. On the other hand, physic and psychologic stressors, provoke changes at CNS and more specifically, at dopaminergic system level.<br/>Given that, Mn and stressful situations affect, directly or indirectly, dopaminergic system function, Mn exposition at the same time that stress exposition could result as an interaction, increasing or modifying the effects of Mn on the CNS. The aim of this thesis is to assess the neurobehavioral effects of Mn prenatal, postnatal and in adults exposure on rodents and interactions of Mn and stress.<br/>Four studies has being done to reach the general goal of this thesis; and in with each one we could assess:<br/> Materno-toxic and feto-toxic effects of hydrocortisone (HC) and Mn prenatal exposure.<br/> Effects of Mn and stress prenatally exposed on postnatal development.<br/> Effects of chronic Mn oral exposure and stress exposure in adults.<br/> Neurobehavioral effects of two kinds of stress: restraint and noise.<br/>All these assessments gave us data about Mn toxicity: maternal, fetal, developmental and in adult animals. We also evaluated two types of stress models because of the rising interest in the assessment of stressful situations and more specifically in order to know the most suitable model of stress to use, in each case, in the future.
