Les metal·lotioneïnes (MT) constitueixen una àmplia família de proteïnes, molt peculiar tant des del punt de vista biològic com des de la perspectiva de la química de coordinació. Es caracteritzen per tenir un baix pes molecular (6-10 kDa) i un elevat contingut en residus de Cys (30%) i en elements metàl·lics, particularment Zn i Cu. Les característiques de les cadenes peptídiques de les MT permeten que aquestes tinguin un paper important en l'emmagatzematge intracel·lular i transport d'ions metàl·lics essencials, així com en la reducció de radicals lliures i d'agents alquilants, i també en el segrestament d'ions metàl·lics tòxics.<br/>L'objectiu aquest treball ha estat determinar la capacitat coordinant de la MT de mamífer, concretament de la isoforma 1 de la MT recombinant de ratolí, enfront d'espècies químiques ben conegudes per la seva toxicitat com Hg(II), MeHg+ i Pb(II).<br/>La metodologia emprada ha consistit a seguir espectropolarimètricament (dicroisme circular) i espectrofotomètrica (absorció UV-Visible) l'estequiometria i les característiques estructurals de les espècies que es formen al llarg de la valoració de Zn7-MT amb les espècies metàl·liques esmentades. D'altra banda, s'ha estudiat el comportament dels fragments a i b de MT per tal d'obtenir dades complementàries que facilitin la interpretació detallada dels resultats de la coordinació dels ions Hg(II), MeHg+ i Pb(II) a la MT sencera. Ha estat necessari analitzar exhaustivament l'efecte de variables, tals com la temperatura, el pH, el temps d'estabilització i el contraió present en solució, en les reaccions de coordinació metàl·lica d'aquests sistemes.<br/>En els resultats obtinguts en les valoracions amb Hg(II), les variables esmentades han estat determinants de la naturalesa de les espècies que es formen. Així, l'estequiometria i l'estructura de les espècies obtingudes a partir de Zn7-MT (pH 7) o d'apo-MT (pH 3), depenen significativament de la força directora de la reacció de complexació (cinètica o termodinàmica) i de la naturalesa dels contraions (Cl- o ClO4-) que acompanyen a les espècies metàl·liques. D'altra banda, la coordinació de Pb(II) a MT dóna lloc a la formació d'una única espècie en un ampli rang de pH (7 - 4,5), l'estructura de la qual depèn del temps d'estabilització. Pel que fa als estudis realitzats sobre el comportament de MT i dels seus fragments a i b enfront de MeHg+ són pioners en l'estudi químic de la coordinació d'aquest ió a aquest tipus de proteïnes.<br/>Tots els resultats obtinguts permeten concloure que la isoforma 1 de MT de mamífer és un bon agent detoxificador de Hg(II), MeHg+ i Pb(II).
Metallothioneins (MT) constitute a wide well-defined group of low-molecular-weight cysteine-rich metal-binding proteins, which are totally atypical under a biological and a coordination chemical viewpoint. Their unusual high cysteine content allows these proteins to be considered responsible for metal homeostasis (Zn, Cu) and detoxification of toxic metal ions (Cd, Hg), free radicals and other electrophilic agents.<br/>The main aim of this work is to analyse the coordination capability of mammalian MT, specifically that of recombinant mouse MT 1, in the presence of highly toxic chemical species such as Hg(II), MeHg+ and Pb(II).<br/>The strategy followed consists of the spectropolarimetric (circular dichroism) and the spectrophotometric (UV-Visible) titrations of Zn7-MT with the above mentioned species together with a detailed analysis of the stoichiometry and the structural features of the successive species formed along the titration. In addition, we have studied the behaviour of the a and b fragments of MT in order to obtain complementary information and thus facilitate the interpretation of the results obtained with the entire protein. A thorough analysis of the effect of some variables, such as temperature, pH, stabilization time and counterions present in solution are fundamental for the study of the metal binding reactions in these systems.<br/>We have shown that the previous variables determine the nature of the species formed along the titration. Thus, the stoichiometry and the structure of the species formed from Zn7-MT (pH 7) or apo-MT (pH 3) are highly dependent on the driving force (kinetic or thermodynamic) of the complexation reaction and strongly influenced by the counterion accompanying the metallic species (Cl- or ClO4-). On the other hand, the coordination of Pb(II) to MT affords formation of a single specie in a wide pH range (7 - 4.5) the structure of which depends on the stabilization time. The study of the behaviour of MT and its corresponding a and b fragments in the presence of MeHg+ is pioneer in the field and reveals that the coordination of MeHg+ to MT entails the unfolding of the protein.<br/>All the results obtained allow conclusion that mammalian MT 1 binds Hg(II), MeHg+ and Pb(II) very efficiently and in this sense it could be used as a heavy metal detoxification agent.
