La disfunció telomèrica com a font d'inestabilitat cromosòmica en cèl·lules epitelials mamàries

Abstract

Els carcinomes usualment presenten inestabilitat cromosòmica (CIN), mostrant tant anomalies estructurals com numèriques. La CIN es genera a l'inici del desenvolupament del càncer i representaria un pas important en l'inici i/o progressió tumoral. Un model per estudiar la CIN deguda a disfunció telomèrica són les cèl·lules epitelials mamaries humanes (HMECs). Quan les HMECs es cultiven in vitro proliferen fins assolir un plateau, anomenat fase de selecció. La inactivació espontània del gen que codifica la proteïna p16INK4a, permet a les cèl·lules seguir proliferant amb una longitud telomèrica per sota de la normal. Els cromosomes amb telòmers disfuncionals poden ser reparats de forma il·legítima conduint a l'aparició de cromosomes dicèntrics els quals són susceptibles d'entrar en cicles de fusió-pont-trencament (BFB) tot generant CIN estructural (S-CIN). <br/>En aquesta tesi es descriuen els mecanismes bàsics de generació CIN deguda a disfunció telomèrica. Primerament, es determinà que l'entrada de cromosomes dicèntrics en cicles BFB genera una CIN massiva que es visualitza amb la presència de morfologies nuclears anòmales (ANM). L'anàlisi de HMECs binucleades demostrà la presència de tres tipus d'ANM: ponts cromatínics, protuberàncies nuclears i micronuclis, els quals incrementaven a mesura que incrementava la disfunció telomèrica. És més, es demostrà que aquestes ANM són originades pels cromosomes amb disfunció telomèrica alhora que s'evidencià un origen comú per als tres tipus d'ANM gràcies a l'estudi de la resolució de ponts anafàsics mitjançant microscòpia a temps real. A més, la utilització de la tècnica de FISH aplicant sondes pancentromèriques i centromèriques específiques per a cromosomes amb escurçament telomèric i cromosomes de longitud telomèrica normal va permetre establir que la disfunció telomèrica a més de generar S¬CIN també pot originar inestabilitat cromosòmica numèrica (W-CIN). Això succeeix quan els ponts anafàsics deguts a disfunció telomèrica segreguen erròniamet durant la mitosi. La capacitat d'un pont anafàsic de no trencar-se durant la divisió cel·lular podria estar condicionada per les forces mecàniques que oposa la cromatina que forma el pont juntament amb la capacitat que tenen els microtúbuls a no perdre la seva unió amb els cinetocors del pont. L'estudi dels ancoratges dels microtúbuls als cinetocors demostrà que els cinetocors dels ponts anafàsics no perden la seva connexió amb els microtúbuls que els uneixen, i poden romandre íntegres més enllà de la sortida de la mitosi. És més, la resistència que oposa la cromatina que intervé en el pont causa un retard en la seva migració fent que se situïn propers a l'equador de la cèl·lula, mentre que altres cromosomes segreguen normalment. Tot plegat, afavoriria la segregació errònia dels ponts anafàsics tot generant aneuploïdies. Per últim, mitjançant citometria de flux es confirmà la presència de HMECs poliploides, l'aparició de les quals està lligada a la interferència dels ponts anafàsics, generats per disfunció telomèrica, amb la consecució de la citocinesi induint la regressió del solc de divisió generant cèl·lules tetraploides. I no només això, sinó que aquestes cèl·lules 4N presentaven nivells més elevats d'inestabilitat cromosòmica, tant estructural com numèrica, que les diploides isogèniques. <br/>Tot plegat, ha permès dibuixar una connexió entre la disfunció telomèrica i l'aparició de tetraploïdies, dos esdeveniments que per separat s'havien relacionat amb la carcinogènesi, però que no havien estat vinculats encara. A l'inici del desenvolupament tumoral la disfunció telomèrica seria responsable de l'aparició de S i W-CIN alhora que també originaria cèl·lules tetraploides. Aquestes cèl·lules tetraploides altament inestables es dividirien de forma asimètrica generant cèl·lules filles aneuploides amb CIN. La posterior reactivació de la telomerasa minimitzaria la CIN deguda a disfunció telomèrica mentre que altres fonts de CIN perdurarien permetent la progressió tumoral.


Epithelial tumours frequently exhibit chromosomal instability (CIN), and show both reorganized chromosomes and aneuploidies. CIN emerges early during carcinogenesis and is an important event in the promotion of tumour development. A valid model for studying telomere-dependent CIN is the human mammary epithelial cell system (HMECs). When these cells are cultured in vitro they proliferate actively until they reach a first plateau phase called selection. Spontaneous inactivation of the gene that codifies for p16INK4a allows cultures to be extended and the generation of CIN due to telomere dysfunction. Chromosomes with dysfunctional telomeres can fuse illegitimately and originate dicentric chromosomes, which may enter into breakage-fusion¬bridge cycles (BFB cycles) that give rise to structural CIN (S-CIN). <br/>Our work describes the different mechanisms through which telomere dysfunction can originate CIN in human cells. We first determined that when dicentric chromosomes enter into BFB cycles, CIN can be visualized by the presence of abnormal nuclear morphologies (ANM). The study of binucleated HMECs showed that there are three main types of ANM, nucleoplasmatic bridges (NPB), nuclear blebs (B) and micronuclei (MN), which increase throughout the cell culture in parallel with increasing telomere dysfunction. Furthermore, using in situ hybridization with fluorescent probes we determined that chromosomes with dysfunctional telomeres were involved in the formation of the different ANM. Real time microscopy revealed that B and MN originated through the breakage of anaphase bridges derived from the fusion of two unprotected chromosomes. Next we wanted to determine if telomere dysfunction was also responsible for the generation of numerical chromosome aberrations in addition to the structural ones. Pancentromeric and centromere specific FISH probes for chromosomes with telomere dysfunction and also with normal telomere length were applied to HMECs. Our results showed that telomere dysfunction not only generates structural CIN (S-CIN) but also whole chromosome instability (W-CIN) due to the missegregation of anaphase bridges during mitosis. The capacity of an anaphase bridge to remain unbroken during mitosis could be related to: 1) the mechanical forces of the intervening chromatin in the bridge that oppose the pulling of kinetochore microtubules at anaphase, and 2) the integrity of the attachment between the kinetochores and the microtubules in the bridged chromatin. The analysis of microtubule-kinetochore attachments showed that chromosome bridges rarely lose their connection with microtubules. In addition, the resistance given by the intervening chromatin in the bridge causes a delay in their migration so that they lag closer to the cell equator, whereas the kinetochores from normal chromosomes are situated close to the poles. Overall, these results imply that bridges that remain near the cell equator could ultimately be missegregated, which would cause aneuploidy. Finally, using flow cytometry approaches it was confirmed that a polyploid HMEC population emerged in the later phases of the culture. These polyploid cells appeared to be related to the presence of anaphase bridges due to telomere dysfunction. Unbroken anaphase bridges trapped below the cleavage furrow ultimately caused cytokinesis regression, which originated binucleated tetraploid cells. Importantly, these polyploid cells were significantly more unstable than the isogenic diploids. In summary, our results showed a connection between telomere dysfunction and the appearance of polyploidy: two events that have been related separately to carcinogenesis in humans. Early in the tumourigenic process telomere dysfunction would originate S and W-CIN as well as polyploid cells. This unstable tetraploid lineage of cells would divide with asymmetric divisions originating a highly aneuploid progeny with CIN. After telomerase activation, CIN, due to telomere dysfunction, would be minimized while other remaining sources of CIN would persist and be responsible for tumour progression.
