En la present Tesi Doctoral s'ha optimitzat i escalat la síntesi del (2R)-2-(1-adamantil)-2-aminoetanol enantiomèricament pur. La seva síntesi parteix de l'acetoacetat d'etil, on s'introdueix el substituent 1-adamantil en la posició intercarbonílica. La posterior transposició d'Schmidt, hidròlisi i reducció ens permet la obtenció del 2-(1-adamantil)-2-aminoetanol de forma racèmica.
La protecció del grup amino del 1,2-aminoalcohol permet la realització d'una ressolució enzimàtica. La Pseudomonas Cepacea transesterifdica més ràpidament l'acetat de vinil per un dels dos enantiòmers del substrat. Controlant la conversió del procés s'acaba obtenint producte de transesterificació amb un 73% e.e. i substracte de partida amb >99% e.e. La posterior desprotecció del grup amino permet obtenir el (2R)-2-(1-adamantil)-2-aminoetanol enantiomèricament pur.
A partir del 1,2-aminoalcohol enantiopur s'ha sintetitzat la (4R)-4-(1-adamantil)oxazolidin-2-ona, es tracta d'una estructura anàloga als magnífics auxiliars quirals que tant bons resultats han donat en síntesi diastereoselectiva. També s'han sintetitzat els lligands quirals bisoxazolina amb simetria C2 2,2'-isopropilidenbis[(4R)-4-(1-adamantil)-2-oxazolina], ((R,R)-Adam-Box), i 2,6-bis[4'-(R)-(1-adamantil)oxazolin-2'-il]piridina, (R,R)-Adam-pybox. Aquests lligands presenten la configuració absoluta invertida respecte als respectius anàlegs comercialment assequibles (S,S)-t-Bu-Box i (S,S)-ip-pybox. Aquests nous lligands sintetitzats presenten una cadena voluminosa i inflexible com a substituent als centres quirals.
El lligand quiral (R,R)-Adam-Box ha estat utilitzat com a lligand de coure (I) en la catàlisi asimètrica de les reaccions de ciclopropanació i oxidació al·lílica. Els valors d'excés enantiomèric obtinguts han estat molt bons (> 98% e.e.) i són comparables als obtinguts amb el seu anàleg comercialment asequible (S,S)-t-Bu-Box.
El lligand quiral (R,R)-Adam-pybox ha estat utilitzat com a lligand de Rodi (I) en la catàlisi asimètrica d'hidrosil·lilació de cetones. En aquest cas no s'obteniren bons resultats.
Ambdós lligands quirals (R,R)-Adam-Box i (R,R)-Adam-pybox, han estat utilitzats com a lligands de coure i metalls lantànids en la catàlisi asimètrica de reaccions de Diels-Alder. S'han obtingut bons resultats de diastereoselectivitat i enantioselectivitat. Aquests mateixos sistemes catalítics han estat usats en la catàlisi asimètrica de diverses reaccions de Michael, s'han obtingut bons rendiments i excessos enantiomèrics. S'han observat uns magnífics efectes del radi iònic del metall i dels impediments estèrics dels reactius en l'enantioselectivitat obtinguda.
Estudis mecanístics realitzats per ressonància magnètica nuclear, espectroscopia UV-Vis i espectrometria de masses, han identificat espècies clau per a la comprensió del mecanisme de la reacció de Michael catalitzada per metalls de transició o lantànids.
S'ha estudiat la capacitat de l'agent de solvatació quiral, (R,R)-ABTE®, com agent enantiodifernciador per ressonància magnètica nuclear de protó de diversos compostos dicarbonílics.

The synthesis of (2R)-2-(1-adamantyl)-2-aminoethanol enantiomericaly pure was achived. Starting from ethyl acetoacetate, 1-adamantyl sustituent was introduced at intercarbonilic position. After Schmidt rearrangement, hidrolysis and reduction racemic 2-(1-adamantyl)-2-aminoethanol was achieved.
Amino proteccion of 1,2-aminoalcohol afforded enzymatic resolution sustrate. Pseudomonas Cepacea transesterificate vinyl acetate faster with one of the sustrate enantiomers, thus, controling process conversion, transesterification product is obtained with 73% e.e. and unreacted stating material is found with >99% e.e. After amino group deprotection, enantiomerically pure (2R)-2-(1-adamantyl)-2-aminoethanol is afforded.
(4R)-4-(1-adamantyl)oxazolydin-2-one was syntethized from the enantiopure 1,2-aminoalcohol, this is a new chiral inductor for diastereoselective synthesis. Bisoxazoline chiral ligands (R,R)-Adam-Box and (R,R)-Adam-pybox were also synthetized from the same enantiopure 1,2-aminoalcohol. These C2-symmetrical chiral ligands present inverted absolute configuration respect their homologous comercially available (S,S)-t-Bu-Box and (S,S)-ip-pybox, and feature a highly steric hindered and inflexible sustituent on chiral carbon.
Chiral ligand (R,R)-Adam-Box was used in copper (I) asymmetric catalysis of cyclopropanation and allylic oxidation reactions. Enantiomeric exceses were good (>98%), comparing results obtained with its homologous commercially available (S,S)-t-Bu-Box.
Chiral ligand (R,R)-Adam-pybox was used in rodium (I) asymmetric catalysis of ketones hydrosilylation. In this case, no good results were obtained.
Both chiral lidands, (R,R)-Adam-Box and (R,R)-Adam-pybox, were tested as chiral inductors when using copper and lanthanides as a metal center on Diels-Alder reaction. Good levels of enantioselectivity and diastereoselectivity were obtained. These catalytic systems were also used on metal-mediated Michael asymmetric catalysis, good yields and enantioselectivities were obtained. A great effect of metal ionic radii and steric hinderance at reagents were observed on obtained product enantioselectivity.
Mechanistic studies based on NMR, UV-Vis and ESI-MS were carried out on metal-mediated Michael catalysis. Some key species were identified for a better iunderstanding of the catalytic process mediated for transition metals or lanthanides.
Chiral solvating agent, (R,R)-ABTE®, was tested as NMR enantiodifferntiating agent for several dicarbonylic compounds.
