Abstract

El principal factor de risc en la malaltia d’Alzheimer, la demència senil més freqüent, és l’edat i presenta major incidència en les dones. En la seva aparició, se li atribueixen també altres factors de risc, tant biològics (genètics i no genètics) com ambientals. La seva forma familiar heretada genèticament (< 5% dels casos), i l’esporàdica (majoritària, d’etiologia desconeguda) difereixen en l’edat d’inici i la taxa de progressió del procés neurodegeneratiu. Tot i això, el fet que comparteixin els mateixos símptomes cognitius i els conductuals i psicològics associats a la demència (BPSD) així com processos neuropatològics subjacents, ha permès a nivell experimental, la utilització de models animals portadors dels transgens humans per a l’avenç en l’estudi de la malaltia. Els ratolins 3xTg-AD (PS1/M146V, APPSwe i tauP301L), creats pel laboratori de Laferla (UCI, USA, 2003), desenvolupen de forma progressiva els trets neuropatològics βA i tau característics de la malaltia, amb un perfil temporal i neuroanatòmic similar al dels pacients. Ells han permès estudiar l’impacte del factor genètic i del gènere (Estudi I) en l’expressió de les conductes bizarres, fins ara no caracteritzades, com a part del seu patró de tipus BPSD primerenc. Alhora, s’han avaluat els efectes beneficiosos a llarg termini del handling postnatal, una estimulació ambiental de tipus sensorial tàctil administrada durant l’ontogènia. El tractament va reduir en els dos genotips, no transgènic (NTg) i 3xTg-AD, la conducta de petrificació i, especialment en les femelles, les conductes bizarres en el camp obert alhora que potencià la conducta de valoració del risc en la caixa fosca/clara. A més, va exercir efectes selectius sobre les ‘estratègies per fer front a l’estrès’ sense modificar l'activitat exploratòria per se. Estudi II. La cafeïna exerceix efectes farmacològics estimulants però també ansiogènics i els efectes d’aquest hàbit de vida són més acusats en els homes. Es va verificar si els beneficis cognitius que se li atribueixen en vers a la malaltia serien extensibles si existeix un perfil BPSD accentuat. Els resultats van demostrar que el tractament oral crònic (dosi baixa, 0,3mg/Kg) de cafeïna iniciat als 6 mesos d’edat (inici de la simptomatologia) va modificar a llarg termini la major part de les conductes als 13 mesos d’edat (inici de les fases avançades). La caracterització d’estratègies en el laberint de Morris van permetre definir patrons d’aprenentatge i memòria distintius (3xTg-AD: natació en cercles i estratègies mixtes i no dirigides; NTg: presència de flotació i estratègies úniques i dirigides) que van ser millorats per la cafeïna. Però, la cafeïna va incrementar el perfil ansiós i agreujar el patró BPSD dels 3xTg-AD, incloent el ritme circadià. Estudi III. Donat que l’edat és el principal factor de risc per a desenvolupar l’AD de tipus esporàdic, es va estudiar un model farmacològic d’envelliment accelerat basat en l’increment de l’estrès oxidatiu induït per l’administració crònica s.c. de 100mg/Kg de D-galactosa. En la soca c57bl/6, els efectes només es van observar a nivell d’equilibri en els mascles i en la coordinació motora en ambdós gèneres. Per contra, la D-galactosa va mostrar efectes positius dosi dependents (0-50-100mg/Kg) en altres dimensions com l’activitat exploratòria, les conductes bizarres així com algunes tasques d’aprenentatge i memòria, el què suggereix un efecte dual protector/deleteri dosi dependent. Annex. Per la seva transcendència translacional en l’estudi del dolor en pacients amb dèficit cognitiu i/o demència, s'inclouen resultats preliminars que indiquen que els ratolins 3xTg-AD mostren la mateixa resposta de la sacsejada de la cua en aigua freda que els animals NTg, i que es veu alentida en tots dos genotips per l’edat.


Age is the main risk factor in Alzheimer's disease (AD), the most common senile dementia, with higher incidence in women. Both biological (genetic and non-genetic) and environmental factors are other risk factors related to its onset. Familial Alzheimer’s disease (FAD) is genetically inherited (<5% of cases) and sporadic (>95% of cases and of unknown etiology) differ in age of onset and rate of progression of the neurodegenerative process. However, both types of AD share the same cognitive and behavioral and psychological symptoms associated to dementia (BPSD) as well as underlying neuropathological processes. Experimentally, this fact has allowed using animal models harbouring human transgenes to progress in the study of the disease. 3xTg-AD mice (PS1/M146V, and APPSwe tauP301L), engineered by Laferla’s lab (ICU, USA, 2003) gradually develop βA and tau neuropathological hallmarks of the disease, in a temporal and neuroanatomical manner similar to that found in AD patients. They have allowed us to study the impact of the genetic load and gender (Study I) in the expression of bizarre behaviors, not characterized yet, as part of their early-like BPSD pattern. Moreover, we have studied the long-term beneficial effects of postnatal handling, an environmental sensorial tactile stimulation administered during the ontogeny. Postnatal handling reduced freezing behavior in both genotypes -non-transgenic (NTg) and 3xTg-AD- and most of the bizarre behaviors -especially in females- in the open field whereas potentiated risk assessment in the dark/light box. This early life treatment also exerted selective effects on coping with stress strategies but did not modify the exploratory activity per se. Study II. Caffeine exerts stimulant pharmacological but also anxiogenic effects. The effects of this habit of life are more pronounced in men. In this study we verified whether the cognitive benefits of caffeine attributed to the disease would be extended in the presence of a strong BPSD profile. The long term effects of a chronic oral treatment (low dose, 0,3mg/kg) of caffeine initiated at 6 months of age (onset of symptoms) were found in most of the behaviors assessed at 13 months age (beginning of the advanced stages). The characterization of strategies in the Morris maze allowed to define distinctive learning and memory patterns (3xTg-AD: circling and mixed and non-goal directed strategies, NTg: floating and single and goal directed strategies) that were improved by caffeine. However, caffeine increased the anxious profile and worsened the BPSD pattern of 3xTg-AD, including circadian rhythm. Study III. Since age is the main risk factor for developing sporadic type of AD, we studied a pharmacological model of accelerated aging based on the increased oxidative stress induced by a chronic administration of D-galactose (100 mg/kg, s.c). In c57bl/6 mice, the aging effects induced by D-galactose were only observed in the equilibrium of males and motor coordination in both genders. In contrast, D-galactose showed positive dose dependent (0-50-100mg/kg) effects in other behavioral domains such as exploratory activity, bizarre behaviors and learning and memory tasks, suggesting a dual and dose dependent protective/deleterious effect. Appendix. Preliminary results have been included in this thesis due to the relevance at the translational level in the study of pain in patients with cognitive impairment and/or dementia. These results indicate that 3xTg-AD mice show the same tail flick response in cold water as compared to NTg animals, which is slowed in both genotypes by age.
