Abstract

La Tesi Doctoral aquí presentada s'ha dedicat a l'estudi del comportament de diferents metal·lotioneïnes de diversos organismes envers els ions metàl·lics Zn(II), Cd(II) i Cu(I), amb la finalitat de validar una nova proposta de classificació d'aquestes proteïnes i a l'hora aprofundir en la seva relació estructura/funció.<br/>Les Metal·lotioneïnes (MTs) són unes proteïnes de baix pes molecular (3-10 kD) amb un elevat contingut en residus de cisteïna, les quals els confereixen la capacitat d'enllaçar metalls pesants. Si bé se les ha relacionat amb diferents funcions dins l'organisme (destoxificació enfront de metalls pesants, homeòstasi de Zn i Cu, funcions antioxidants i antiapoptòtiques, etc), encara es treballa intensament per determinar la seva funcionalitat específica. Aquest fet ha dificultat la seva classificació, de manera que fins a l'actualitat s'han fet diferents propostes. La més recent es centra en la preferència metàl·lica de les MTs envers els ions essencial Zn(II) i Cu(I). Aquests ions, amb comportaments coordinants clarament diferenciats, probablement determinen estructures diferents per als complexos M-MT, les quals, tenint en compte la relació estructura/funció, segurament porten a funcions diferenciades per a les anomenades Zn- i Cu-tioneïnes.<br/>En aquesta Tesi Doctoral es recullen els estudis de la química de coordinació de: la última isoforma de MT de mamífer descoberta, la MT4; d'una MT d'un organisme evolutivament proper als mamífers, en concret la ckMT de pollastre, i d'una MT d'un organisme evolutivament més allunyat a mamífer, en particular la Crs5 de llevat. Addicionalment, s'inclouen els resultats obtinguts durant l'estudi d'aquestes i d'altres MTs d'organismes diferents en presència de lligands no proteics (clorurs i/o sulfurs) en els complexos metall-MT. <br/>Els resultats obtinguts en l'estudi de MT4 han permès classificar aquesta MT com una Cu-tioneïna (en contraposició a MT1 que havia estat prèviament classificada com a Zn-tioneïna), de tal manera que MT4 esdevé la primera Cu-tioneïna trobada en organismes superiors. A més, s'ha pogut determinar que les diferències de comportament entre MT4 i MT1 enfront de Cd(II) són degudes als respectius fragments ? (?MT1 i ?MT4), mentre que les seves diferències en l'enllaç a Cu(I) s'han atribuir als respectius fragments ? (?MT1 i ?MT4). També s'ha proposat que MT4 proporciona un entorn de coordinació "particular" per al Cu(I) que MT1 no pot oferir. Així mateix, l'anàlisi espectroscòpica detallada dels camins de reacció de MT1 i MT4 envers Zn(II) i Cd(II) ha proporcionat evidències de la contribució de lligands clorur (Cl-) en l'estabilització de l'estructura dels complexos Cd-MT1 però no en la dels complexos Cd-MT4, Zn-MT1 i Zn-MT4. Totes aquestes característiques diferencials entre MT1 i MT4 suggereixen que probablement MT4 i MT1 presenten funcions fisiològiques diferents en mamífer.<br/>Els estudis realitzats amb ckMT han portat a classificar-la com una Zn-tioneïna, però alhora també han permès proposar l'existència d'una gradació entre el caràcter de Zn- i Cu-tioneïna de les MTs. Així, ckMT presenta una capacitat coordinant envers Zn(II), Cd(II) i Cu(I) intermèdia entre les descrites per a MT1 i MT4, amb un comportament envers els metalls divalents més proper al de MT1, però més similar al de MT4 quan enllaça Cu(I). La presència d'un residu de His en l'extrem C-terminal de ckMT li confereix capacitats coordinants especials enfront de Cd(II) que la diferencien de MT1 i MT4. Aquesta His no només coordina Cd(II), sinó que és la responsable de la formació de dímers mitjançant un procés de dimerització mai descrit fins al moment per a MTs.<br/>Els resultats obtinguts en l'estudi de Crs5 de llevat han corroborat la gradació entre Zn- i Cu-tioneïnes proposada en el cas de ckMT, de manera que Crs5 podria comportar-se com una Zn-tioneïna en situacions de baixes concentracions de Cu intracel·lular, mentre que actuaria com una Cu-tioneïna en presència de concentracions de Cu significatives. Un altre fet interessant és que en la síntesi recombinant de Crs5 el grau d'oxigenació dels medis de cultiu enriquits en Cu afecta la naturalesa homo- o heteronuclear dels agregats M-Crs5 que se'n recuperen, on M = Zn(II) i/o Cu(I), obtenint-se espècies homometàl·liques Cu-Crs5 si no s'oxigena el medi i complexos heterometàl·lics Zn,Cu-Crs5 per oxigenacions elevades.<br/>Molts dels resultats obtinguts en l'estudi de les MTs objecte d'aquesta Tesi així com en els de MTs d'altres organismes han proporcionat al grup de recerca en el marc del qual s'ha realitzat aquest treball indicis de la participació d'un altre lligand no proteic en els complexos M-MT (M = Zn i/o Cd): el lligand sulfur (S2-). Així, com a part d'aquest treball, també s'han dut a terme un seguit d'experiments mitjançant els quals s'ha pogut determinar que totes les espècies recombinants Zn- i Cd-MT estudiades contenen lligands sulfur, obrint així noves i prometedores perspectives, tant estructurals com funcionals, en el camp de les MTs.


The following Thesis has been devoted to the study of the Zn(II), Cd(II) and Cu(I) coordinative abilities of different metallothioneins (MTs) from several organisms, with the purpose of validating a new proposal for MT classification and deepening further into the structure/function relationship of these proteins.<br/>MTs are low molecular weight proteins (3-10 kD) with a high cysteine content, which confers them the ability to bind heavy metal ions. Although they have been related to different functions in the organism (heavy metal detoxification processes, Zn and Cu homeostasis, antioxidant and antiapoptotic functions, etc), their physiological function or functions is still a matter of debate and prompts most of the research within this field. This fact has difficultated their classification, and in consequence several models of classification have been proposed. The most recent one is based on the metallic preferences of MTs towards the essential Zn(II) and Cu(I) ions. These ions clearly show different coordinative abilities and must therefore determinate the formation of different M-MT structures. Therefore, when considering the structural/function relationship, these differences in the structure of the M-MT complexes might determine different functions for the so-called Zn- and Cu-thioneins.<br/>This work gathers the coordinative abilities of the last mammalian MT isoform discovered (MT4), one MT from an organism evolutionarily close to mammals, which is chicken (ckMT1), and one MT from an evolutionarily distant organism, yeast (Crs5). Additionally, the study of the binding properties of these MTs and others from other organisms has been performed in the presence of non-proteic ligands (chloride and/or sulphide).<br/>The results obtained during the study of MT4 have proved that this isoform is a Cu-thionein (in contrast with MT1, which was previously classified as a Zn-thionein), and therefore MT4 is the first Cu-thionein ever reported in high organisms. Furthermore, it has been proved that the differences in Cd(II) coordination between MT4 and MT1 should be assigned to their ? domains (?MT1 and ?MT4), whereas the ? domains (?MT1 and ?MT4) are the responsible for the differences in Cu(I) coordination. It has also been proposed that Cu(I) is bound to MT4 through a "particular" coordination environment that MT1 cannot offer. Moreover, a detailed spectroscopic analysis of the reaction pathways of MT1 and MT4 towards Zn(II) and Cd(II) has gathered evidence for the contribution of chloride (Cl-) ligands in the stabilization of the structures of the Cd-MT1 species, but not in the Cd-MT4, Zn-MT1 and Zn-MT4 species. These differences between MT1 and MT4 isoforms suggest that both isoforms are probably involved in different physiological processes in mammals.<br/>The studies performed with chicken MT have shown that ckMT1 exhibits a Zn-thionein behaviour, although there seems to be a gradation between the Zn- and Cu-thionein character in MTs. Thus, the Zn(II), Cd(II) and Cu(I) binding abilities of ckMT are intermediate between those of MT1 and MT4, with a divalent metal ion behaviour closer to MT1, but more similar to MT4 when binding Cu(I). The presence of a His residue at the C-terminal position of its ? domain confers to this protein some special Cd(II) binding abilities which are significantly different from those of MT1 and MT4. This His residue does not only bind to Cd(II) but also is the responsible for dimer formation though a process that has never been described before for MTs.<br/>The results obtained with yeast MT (Crs5) have corroborated the gradation between the Zn- and Cu-thionein character already proposed in the study of ckMT1 in such a way that Crs5 could behave as a Zn-thionein when intracellular Cu concentration is low, whereas in the presence of significant Cu amounts in the cell it would behave as a Cu-thionein. Another interesting point relies on the fact that during the recombinant synthesis of Crs5 the amount of dissolved oxygen in the Cu enriched media affects the homo- or heteronuclear nature of the M-Crs5 complexes that are recovered, where M = Zn(II) and/or Cu(I): when the cultures are not oxygenated at all only Cu-Crs5 homometallic species are obtained, whereas Zn,Cu-Crs5 heterometallic complexes are obtained for high oxygenation levels.<br/>Many of the results obtained here as well as the results from other people in the same research group working with MTs from other species have shown signs for the participation of another non-proteic ligand in the M-MT species (M = Zn and/or Cd): the sulphide ligand (S2-). Therefore, several experiments have been performed during this PhD that have permitted to prove the presence of sulphide ligands in all the recombinant Zn- and Cd-MT species here analysed. This discovery opens promising new structural and functional possibilities for these special proteins.
